Harvested chromatin was sonicated using the Bioruptor UCD-200 for 2 cycles of 10 min (max power 30 s on 30 s off). After sonication, chromatin was incubated with designated amount of antibody at 4 °C overnight. Immunoprecipitated complexes were collected by Protein A/G Magnetic beads (Pierce, 8802). Sample integrity and purity were detected by Agilent Technologies 2100 bioanalyzer. DNA was 3' adenylated and PCR amplified. dsDNA products were heat denatured and circularized by splint oligo for formating the ssCircularDNA library.
