Cells were washed in phosphate-buffered saline (PBS), cross-linked with DSG for 45 minutes followed by fixation with 1% formaldehyde (methanol-free) for 10 minutes at room temperature, quenched with 0.125 M glycine for 5 min at room temperature and washed with PBS complete. Cells were resuspended in lysis buffer (1%SDS, 50 mM Tris-HCl pH8, 10 mM EDTA) containing protease and deacetylase inhibitors, and incubated on ice for 15 min. Lysed cells were sonicated for 5 minutes with a Covaris E220 sonicator (PIP140, DF5%, 200cpb), in 1ml AFA Fiber milliTUBEs. 5 µg of soluble chromatin were immunoprecipitated overnight with 10 μg of c-JUN antibody (CST 9165) on protein A and G magnetic beads; and finally washed, eluted and DNA purified. ChIP-seq libraries were constructed using Accel-NGS 2S DNA library kit from Swift Biosciences for c-Jun ChIP-seq.
