Supporting cells for the developing female gamete in the OVARY. They are derived from the coelomic epithelial cells of the gonadal ridge. Granulosa cells form a single layer around the OOCYTE in the primordial ovarian follicle and advance to form a multilayered cumulus oophorus surrounding the OVUM in the Graafian follicle. The major functions of granulosa cells include the production of steroids and LH receptors (RECEPTORS, LH).

Granulosa cells were collected from 30 mice and plated in six petri dishes (each with at least 1×107 cells). Chromatin was isolated using the ChromaFlash Chromatin Extraction Kit (EpiGentek, Cat. #P-2001) and sheared using the EpiSonic 2000 Sonication System (EpiGentek, Cat. #EQC-2000). DNA fragment size distribution was determined to be 100 to 300 bp by Agilent Bioanalyzer analysis. The concentration of DNA following ChIP was between 3.5-16.3 ng. Library Preparation was done by DNA end polishing and adaptor ligation, library amplification using indexed primers and library purification. Library was verified on a Bioanalyzer and KAPA qPCR Library Quantification.RNA sequencing libraries were prepared using 500 ng RNA, and the NEB Next Ultra RNA Library Prep Kit for Illumina using the manufacturer's instructions (NEB, Ipswich, MA, USA)
