A subset of helper-inducer T-lymphocytes which synthesize and secrete INTERLEUKIN-2; INTERFERON-GAMMA; and INTERLEUKIN-12. Due to their ability to kill antigen-presenting cells and their lymphokine-mediated effector activity, Th1 cells are associated with vigorous delayed-type hypersensitivity reactions.

For ChIP-seq, 10 million Th1 cells overexpressing Trp73 or vector control cells were performed using anti-FLAG (M2) antibody (Sigma) as previously described. For RNA-seq, Trp73 overexpressed Th1 cells were isolated by FACS sorting as GFP+ cells three days after retroviral transduction, and total RNA was then extracted. RNA-Seq libraries were prepared from mRNA (isolated from 2 μg total RNA) using the KAPA kit per the manufacturer's protocol. After quantification by Qubit (Invitrogen), barcoded samples were mixed and sequenced on an Illumina HiSeq2000 or HiSeq3000 system.
