FFPE blocks were histologically examined and sectioned. Samples were deparaffinized, rehydrated and homogenized by heating at 50C, 65C or 80C in 1% SDS detergent buffer overnight. Next samples were sonicated for either 5 or 40 minutes and solubilized chromatin was immunoprecipitated following a low-input protocol. The optimal condition (65C and 5 minute sonication) was done in replicate. Libraries were prepared with Accel-NGS 2S Plus DNA Library Kit (Swift Biosciences) or SMARTer® ThruPLEX® DNA-Seq Kit (Takara Bio USA Inc).
