Following fixation, Nac punches were sonicated with a desktop sonicator and then underwent high power sonication in a Bioruptor for 30 cycles of 30 s on/30 sec of. The machine was cooled before one more round of 10 cycles. Beta-catenin AB (ChIP validated, Cell Signaling 4176) was used for IP. IPed DNA was reverse cross-linked at 65 degrees overnight and purified. Each replicate was 15 mice pooled, 4 14 gauge NAc punches/animal Libraries were prepared for sequencing with standard Illumina Protocol. 200-400 bp fragments were selected for sequencing.
