Finely minced liver tissue was incubated in PBS containing 1% formaldehyde for 10 min, and then, glycine was added.The samples were resuspended in hypotonic buffer and lysed by homogenization. Nuclei were pelleted and sonicated. The chromatin sample was precleared and immunoprecipitated by binding to Protein G agarose with the H3K27-me3 (Abcam) antibody, the beads were washed, chromatin was eluted, and DNA was isolated. ChIP assays were performaed using H3K27-me3 antibody (Ab6002) . Two sets of input and immunoprecipitated samples from 2 mices were used respectively. Libraries were prepared accoding to standard Illumina protocols.
