30,000 to 75,000 bone marrow CD45+CD11b+Ly6ChiLy6G– monocytes were flow sorted (pools of 2 mice), then washed in cold PBS and lysed for 1 min in a refrigerated centrifuge. Transposition reaction (Illumina) was performed at 37°C for 30 min. After purification of the DNA with the MinElute PCR purification kit (Qiagen), material was amplified for 5 cycles. Additional PCR cycles were evaluated by real time PCR. Final product was cleaned with the MinElute PCR purification kit. Libraries were individually checked with the Kapa-Roche quantification kit and size was verified on tapestation high sensitivity screentape.
