Single-cell thymocyte suspensions were acquired with mechanical dissociation and cells were stained with appropriate antibodies and tetramers. Samples were flow-sorted in PBS with 0.5% FBS and were then processed for ATAC-seq. Library was constructed according to standard Illumina protocols
