Lymphocytes from the colonic and small intestine LP were isolated using the Lamina Propria Dissociation Kit (Miltenyi Biotec) according to the manufacturer's protocol. ILCs were enriched using the Pan-ILC enrichement kit (Stem Cell Technologies), and 20,000-30,000 ILC1s were FACS sorted as Lineage(TCRβ, TCRγδ, CD8α, CD8β, CD19, CD11b, DX5, Gr-1, Ter119)-Thy1.2+CD127+NK1.1+NKp46+. ILC1s were lysed and transposase reaction was performed with Tn5 transposase (Illumina) at for 30 min at 37C. DNA was purified using the MinElute kit (Qiagen). Libraries were amplified using Nextara primers with NEBNext High-Fidelity 2X PCR Master Mix (New England Biolegends), and reactions were purified with the PCR Purification Kit (QIAGEN).
