Epidermal cells which synthesize keratin and undergo characteristic changes as they move upward from the basal layers of the epidermis to the cornified (horny) layer of the skin. Successive stages of differentiation of the keratinocytes forming the epidermal layers are basal cell, spinous or prickle cell, and the granular cell.

DNA was extracted by incubating the cells in 50ul of cold lysis buffer (10 mM TRIS-HCl, pH 7.4, 10mM NaCl, 3 mM MgCl2, 0.1% Igepal CA-630) for 15 min while centrifuging (500 x g, 4°C) Pelleted nuclei were resuspended in transposition reaction mix with Tn5 Transposase (Illumina FC-121-1030) and incubated 45 min with gentle shaking at 37°C. Tagmented DNA was purified using SPRIselect beads (Beckman Coulter B23317) and amplified by PCR. Resulting libraries were size-selected and purified again by SPRIselect beads
