Main types are PERITONEAL MACROPHAGES; ALVEOLAR MACROPHAGES; HISTIOCYTES; KUPFFER CELLS of the liver; and OSTEOCLASTS. They may further differentiate within chronic inflammatory lesions to EPITHELIOID CELLS or may fuse to form FOREIGN BODY GIANT CELLS or LANGHANS GIANT CELLS. DNA was isolated as previously described (Buenrostro, J. D., Giresi, P. G., Zaba, L. C., Chang, H. Y. & Greenleaf, W. J. Transposition of native chromatin for fast and sensitive epigenomic profiling of open chromatin, DNA-binding proteins and nucleosome position. Nat. Methods 10, 1213–1218, 2013). 50,000 cells were used for the transposition reaction, that was carried out for 4h at 37°C. Library fragments were amplified using 1× NEBnext PCR master mix and 1.25 μM of custom Nextera PCR primers. using the following PCR conditions: 72 °C for 5 min; 98 °C for 30 s; and thermocycling at 98 °C for 10 s, 63 °C for 30 s and 72 °C for 1 min - this was done for 13 cycles. Transposed DNA fragments were then PCR amplified for 13 cycles and purified by the ChargeSwitch PCR clean-up kit (Invitrogen)
