Lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody. To generate libraries for high-throughput sequencing, purified DNA fragments were end-repaired, adenylated, ligated to adaptors and PCR amplified for 18 cycles. The PCR products corresponding to 250-450 bps were gel purified, quantified and stored at -80℃ until used for sequencing.For high-throughput sequencing, the libraries were prepared following the manufacturer's instructions and applied to illumina NextSeq 500 system for 151 nt pair-end sequencing
