Mice were immunized with MOG as described above. TCRb+YFP+ cells were sorted from dLN or spleen at day 9 post-immunization Cells were lysed in 50 ml ATAC-Seq lysis buffer (10 mM Tris-HCl, pH 7.4, 10 mM NaCl, 3 mM MgCl2, 0.1% IGEPAL CA-630) on ice for 10 min. Resulting nuclei were pelleted at 500 g for 10 min at 4°C. Supernatant was carefully removed with a pipette and discarded. The pellet was resuspended in 50 ml transposase reaction mix (25 ml 2× TD buffer, 22.5 ml nuclease-free water, 2.5 ml Transposase) and incubated for 30 min at 37°C
