Inguinal and perigonadal adipose was dissected and finely minced. Tissue was subjected to cross linking in disuccinimidyl glutarate for 30 minutes followed by 1% formaldehyde for 10 minutes. Reaction was quenched with glycine. Tissue was dounced to release nuclei. Sun1 positive nuclei from adipocytes were collected by addition of c-myc magnetic beads. Enriched nuclei were sonicated to shear the chromatin and subjected to immunoprecipitation. Precipitates were washed and DNA was purified by column (Qiagen Min-Elute). KAPA Hyper Prep Kit (Kapa Biosciences)
