ATAC-seq was performed as previously described (Buenrostro et al., 2013) with minor adjustment for application to tissue material. Six fresh L4-L6 DRGs were extracted in 100 µl ice cold cell lysis buffer (10mM Tris-HCl pH 7.4, 10mM NaCl, 3mM MgCl2, 0.5% IGEPAL CA-630) for 5 minutes on ice, then a 21g needle on a 1 mL syringe was used to shear the tissue through the needle 5 times.
