They may further differentiate within chronic inflammatory lesions to EPITHELIOID CELLS or may fuse to form FOREIGN BODY GIANT CELLS or LANGHANS GIANT CELLS. Chromatin was digested with MNase, enriched with indicated antibodies, and purified with QIAquick PCR purification kit (Qiagen, #28106). Total RNAs were prepared by following protocols from RNeasy Plus Mini Kit (Qiagen, 74134). ChIP-seq libraries were constructed with Accel-NGS 2S Plus DNA Library Kit (Swift Biosciences) according to the manufacturer's protocol. The libraries were then amplified and assessed for fragment size using TapeStation (Agilent) and quantified using Qubit dsDNA HS Assay Kit (Thermo Fisher Scientific). RNA-seq libraries were constructed with Illumina TruSeq Stranded mRNA Library Prep Kit Set A (Illumina, RS- 122-2101). The libraries' size was selected by using the Agencourt AMPure XP beads (Beckman Coulter, A63882), with average size 400bps.
