Lysates were clarified from sonicated nuclei and transcription factor PBX1-DNA complexes were isolated with antibody. A total amount of 10ng qualified ChIP-ed DNA per sample was used as input material for the library preparation. The sequencing libraries were generated using the VAHTS Universal DNA Library Prep Kit for Illumina® (Vazyme, ND604) following manufacturer's recommendations. Firstly, the end repair of DNA fragments was performed and an "A" base was added at the 3'-end of each strand. Adapters (Vazyme, N801) were then ligated to both ends of the end repaired/dA tailed DNA fragments for amplification and sequencing. The DNA fragments were purified and selected with VAHTSTM DNA Clean Beads (Vazyme, N411) in order to get appropriate size for sequencing. PCR was performed and aimed products were purified finally.
