Cell lines before ChIP were grown in White Medium DMEM with CDT. Cells expressing the Y537S mutation were grown in Doxycyclin.The cells with WT-ER were stimulated with E2 x45 minutes prior to cross linking for the ChIP experiment. Cells were sonicated, then cell lysate was incubated with antibody to extract protein-DNA complex of interest, Library Preparation is performed using the Rubicon 48S kit
