Nuclei were isolated and chromatin was sonicated and precleared. The H2A.Z or H3 histone-DNA complexes were isolated using antibody against H2A.Z (Abcam, ab4174) or using antibody against H3 (Abcam, ab1791) . The immunoprecipitated DNA was then purified and 10 ng was sent to BGI (HONG KONG) or EMBL GeneCore (Heidelberg, Germany) to perform library construction and sequencing. Library construction and sequencing was performed by BGI TECH SOLUTIONS (HONGKONG) for the first replicates of H2A.Z ChIP-Seq in proliferation and in senescence, and at EMBL GeneCore for the second replicates of H2A.Z ChIP-Seq in proliferation and in senescence as well as for the H3 ChIP-Seq in proliferation and in senescence.
