Lysates were clarified from sonicated nuclei with Covaris M220 and histone-DNA complexes were isolated with antibody. Libraries were prepared using NEBNext Ultra DNA Library preparation kits from 2-10 ng immunoprecipitated DNA according to manufacturer’s protocol. The finished double strand DNA (dsDNA) libraries were quantified by PicoGreen and Agilent Bioanalyzer according to manufacturer’s protocols. Uniquely indexed libraries were pooled in equimolar ratios and sequenced on an Illumina NextSeq500 with single or paired-end 75bp reads.
