Lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody using Easy Magna Chip kit (Catalog # 17¬10085 & # 17¬10086, Millipore Sequencing libraries were generated using library Preparation Kit (NEB) following manufacturer’s recommendations and index codes were added to each sample. Briefly, the DNA sample was fragmented by sonication to a size of 75~100bp, and then DNA fragments were end polished, A-tailed, and ligated with the full-length adapter for Illumina sequencing with further PCR amplification.
