Cells were pelleted, washed once in PBS, resuspended in lysis buffer(10mM Tris.HCl pH7.4, 10mM NaCl, 3mM MgCl2, 0.1% v/v Igepal CA-630) and the nuclei pelleted at 4 degrees celsius. Transposase reaction and library preparation was then performed using the Illumina Nextera DNA Library prep kit (Cat. No. FC-121-1030) exactly as described in Buenrostro et al. 2015 (Curr. Protoc. Mol. Biol. 109:21.29.1-21.29.9) and four libraries were multiplexed per lane on a HiSeq 4000.
