Heterozygous Nestin-GFP mice were bred with homozygous DCX-mRFP mice to yield GFP/RFP double positive embryos. The cortices derived from the double positive embryos were dissociated by trituration in HBSS with 2mM of EDTA and 25 ug/ml of DNase I. Cells were washed once with HBSS and re-suspended in DMEM/F12. Fluorescence-activated cell sorting (FACS) was used to acquire neural progenitor cells (NPCs, green color) and neurons (double color). Lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody. DNA samples after ChIP were subjected to amplification with MicroPlex Library Preparation kit (AB-004-0012, Diagenode) according to the instruction.
