Cells were lysed and the chromatin was sheared to a size range of 200 to 400 bp using Covaris M220 sonicator. Sheared chromatin was diluted and incubated at 4°C overnight with antibodies against Histone H3 trimethyl Lys4 (K4, Millipore #07-473), Histone H3 trimethyl Lys27. ChIP samples were prepared and indexed for Illumina sequencing using the TruSeq DNA sample Prep Kit.
