10,000 HSPCs were sorted into PBS buffer and cross-linked with 1% formaldehyde at RT for 10 min, and 0.125 M glycine was added to stop the reaction at RT for 5 min, followed by washing once with ice-cold Hank's buffered salt solution (HBSS) containing protease inhibitor cock¬tail (PIC). Libraries were constructed with the KAPA library preparation kit for Illumina (Kapa Biosystems).
