Intact nuclei were isolated from M1 and IL-6 treated M1 cells and restrictedly digested with DNase I according to a standard protocol (Sabo et al., 2006). Fractions containing small DNA fragments representing two-hit DNase I cutting events were precipitated, pooled, and prepared for Illumia sequencing according to the manufacturer Illumina.
