Cells were fixed with 0.7% formaldehyde for 10 min, lysed, and sonicated using a Bioruptor (Diagenode). Approximately 1% of sonicated nuclear lysate was removed to serve as an input control. Sonicated nuclear lysate was incubated with ARNT antibody overnight, and the next day, immunoprecipitated chromatin fragments were captured using protein G-conjugated Sepharose beads. Control (input) and immunoprecipitated DNA fragments were then eluted and purified. Libraries were generated using the NEBNext Ultra II DNA Library Prep Kit (New England Biolabs).
