DNA was isolated from c-Kit+ splenocytes or single cell suspensions from CFU using the Qiagen Puregene kit according to manufacturer’s instructions. For small cell numbers glycogen was added to help precipitate DNA. Enhanced Reduced Representation Bisulfite Sequencing (ERRBS) libraries were generated from 25 ng of genomic DNA as previously described (Akalin et al., 2012) for the human specimens, and sequenced on a HiSeq-2000 sequencer. Murine libraries were prepared in the same way but using the indexed adaptors and sequenced with two libraries per lane on a HiSeq-2500 sequencer (Garrett-Bakelman et al., 2015).
