ChIP for H3K4me1 (Abcam ab8895) was performed using sonicated extracts from formaldehyde-fixed nuclei. HeLa S3 cells were grown in DMEM + 10% foetal calf serum (FCS) Untreated DNA was extracted from immunoprecipitated or input chromatin using Qiagen MinElute kit and re-sonicated to obtain a mean fragment size of around 300bp. Resonicated DNA was ligated to NEBNext adapters using the NEBNext Ultra II DNA Library Prep Kit (New England Biolabs), and amplified by PCR (8 cycles).
