Sertoli cells were isolated from testes of 6 dpp mice and maintained in DMEM/F12 media supplemented with 2.5%FBS, 5%horse serum, 2mML-Gln, 100U/mLpenicillin, 1uMsodium pyruvate Sertoli cells lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody. Cells were cross-linked with 1% formaldehyde for 10 min at room temperature and formaldehyde was then inactivated by the addition of 125 mM glycine for 5min. Sonicated DNA fragments with 100–300 bp were pre-cleared and then immunoprecipitated with Protein A+G Magnetic beads coupled with Anti-Androgen Receptor antibody (06–680, Millipore).
