The testes were crosslinked with 1% formaldehyde treatment and fixation was stopped by addition of 0.125 mM glycine. After washing with PBS, samples were stored at -80 C. The crosslinked material from 100 mg testes was sonicated to 500 bp fragments by Covaris S2 (duty cycle; 20 %, cycle per burst; 200, 10 times for 30 seconds). Immunoprecipitation was performed by incubatin overnight at 4 C with 2 ug of anti-H3K27me3 (Millipore 07-449), 2 ug of anti-H3K4me1 (abcam ab8895), 4 ug of anti-H3K4me3 (abcam ab1012), and control IgG. Antibody-protein complex was captured with dynabeads protein A or G beads (life technologies) and DNA was extracted.
