600 starved L1 larvae born at room temperature (21° C - 22° C) overnight in M9 Buffer were collected, frozen in liquid nitrogen, and stored at -80° C prior to homogenization. Frozen pellets were disrupted by a glass Dounce homogenizer, fixed with formaldehyde (1% final concentration), and quenched with glycine. ChIP samples were processed with a Chromatin Immunoprecipitation Assay Kit (Millipore), according to manufacturer's instructions. Samples were sonicated using a Diagenode Bioruptor UCD-200 at 4 °C on the “high” setting for a total of 30 min with a cycle of 45 secs on and 15 secs off. A total of 12.5 μL (5 ug) H3K36me3 antibody (cat. 61021; Active motif) was used for immunoprecipitation. The Genomic Services Laboratory (GSL) (HudsonAlpha, Huntsville, Alabama) performed library preparation and sequencing (Illumina HiSeq v4, 50bp single-end reads). Libraries were prepared by HAIB-GSL (for HiSeq).
