Cells were fixed in 1% formaldehyde for 15 min at room temperature. Crosslinking reaction was extinguished by adding glycine to 125 mM. Cells were washed three times in PBS, 0.5% lgepal CA-630, 1 mM phenylmethanesulfonyl fluoride (PMSF). Cell pellets were snap frozen on dry ice and stored at -80°C ChIP-Seq was performed by Active Motif (Carlsbad, CA). Chromatin (30 ug) from vehicle or ACY-957 treated cells was immunoprecipitated with antibodies against Gata-2 (20 ul, sc-9008, Santa Cruz, Dallas, TX), HDAC1 (20 ug, ab7028, Abcam, Cambridge, MA), or HDAC2 (4 ug, ab12169, Abcam). ChIP DNA, as well as a control sample composed of equal amounts of input DNA from each treatment group, was processed into Illumina libraries using standard procedures, and libraries were sequenced on HiSeq 2000.
