Testes were harvested in Trizol for RNA-Seq, and fixed with formaldehyde for ChIP-Seq Rabbit polyclonal antibodies to DMRT6 were raised against a purified DMRT6 fusion protein containing glutathione-S-transferase (GST) fused to the C-terminal 148 amino acids of DMRT6. Antibodies to GST were removed by GST-affigel 10 chromatography and the antiserum was then purified by GST-DMRT6-affigel 10 chromatography RNA libraries were made using Illumina TruSeq Protocol. The ChIP libraries were made from the input and the material immuniprecipitated with the Dmrt6 antibody described in this paper. Chromatin fragments were blunted with Lucigen DNA terminator enyme mix (Lucigen# 40035). Blunted fragments were adenylated with Exo- Klenow (NEB #MO2125). Illumina adapters (Illumina #100521) were diluted 1:50 and ligated to adenylated fragments with T4 ligase (NEB #NEB M0202T). Products were amplified with 18 rounds of PCR using Phusion Polymerase (NEB M0530s) using barcoded oligonucleotides. DNA fragments between 150 and 250 were purified by agarose gel electrophoresis.
