Chromatin was extracted from cross-linked cells, and sonicated to 200-700 bp fragments. Immunoprecipitation was performed with 15 µg of chromatin for 2.5 µg of anti-H3K27ac and H3K9me3 antibody or 75µg chromatin for 5µg Cas9 antibody. DNA was purified using the GeneJET PCR purification kit (Thermo Fisher). Libraries were prepared from 1-5 ng of DNA using the NEBNext ultra II DNA library prep kit.
