For STAT3 ChIP-seq, formaldehyde-fixed human Th1 cells were sonicated using sonicator (Vibra Cell, SONICS) and resulting chromatin fragments equivalent to 10 million cells were isolated by centrifugation and STAT3 bound chromatin complexes were isolated with the anti-STAT3 antibody. For RNA-seq experiments, Total RNA isolated from human Th1 cells that were either not stimulated or stimulated with different IL-6 variants for 6 hours. ChIP-Seq libraries were prepared using the KAPA LTP library preparation kit (Roche Sequencing) and the NextFlex DNA barcode adoptors (Bioo Scientific, PerkinElmer) for multiplexing and libraries were then sequenced on Illumina Hi-Seq 4000 platform (GeneWiz).
