10 x106 cells were fixed with 1% formaldehyde, lysed and sonicated leading to a DNA size of approximately 200bp. Tri-methyl histone H3 (Lys 27) (H3K27me3) monoclonal antibody (Cell Signaling, 9733) was added to the samples and incubated overnight at 4°C. The complex was purified using magnetic beads followed by elution from the beads and de-crosslinking. DNA was purified using PCR purification columns. ChIP sequencing libraries on purified DNA were generated and sequenced by Genewiz (New Jersey, USA)
