We fixed 10-20 million CD4 T lymphocytes, after 4h treatment with 20 mM UNC4976 (0.2% DMSO) or 0.2% DMSO, with 1% formaldehyde for 12 minutes. After fixation, formaldehyde was quenched by adding glycine to a final concentration of 100 mM. The Fixed cells were washed in ice cold PBS, pelleted at 1200g and after decanting remaining PBS, the pellets were flash frozen with liquid nitrogen. After harvesting fixed nuclei and chromatin was sheared and immunoprecipitated as previously described (Hodges and Stanton et al Nature Struct Mol Biol 2017). For these ChIP studies, rabbit anti-CBX7 antibody (Abcam cat# 21873, Lot number: GR255463-2) was used. After reverse crosslinking, ChIP DNA was phenol-chloroform extracted.
