For single fixation with formaldehyde, cells were fixed with 1% formaldehyde-containing medium for 10 min. For double fixation, the cells were first fixed with 2 mM EGS for 30 min followed by with 1% formaldehyde-containing medium for 10 min. Libraries were prepared using KAPA Hyperprep Kit.
