ChIP, ChIP libraries preparation, and deep sequencing (ChIP-seq): Homogenize WT and Taf7l KO testis tissue from 10 3-month-old mice followed by fixation with 1% formaldehyde for 10 min at room temperature then use 0.125 M glycine to stop the crosslinking for an additional 5 min and collect tissue by spinning down. The chromatin obtained from WT and Taf7l KO testis tissue was fragmented to sizes ranging from 175 to 225 bp using a Covaris-S2 sonicator (Covaris, Inc., Woburn, MA) for a total processing time of 20 min (30 s on, 30 s off). 1mg of the sonicated chromatin was used in each immunoprecipitation reaction with 4ug of Pol II and TAF7L antibodies, 8ug equally mixed mouse and rabbit IgG were used as negative control.Preparation of the sequencing libraries on the DNA samples of the immunoprecipitation from antibodies and IgGs precisely followed the instructions from Illumina (Illumina Inc., San Diego, CA).
