Formaldehyde crosslinked chromatin was sonicated to fragments DNA to about 200-300 bp, the immunoprecipitated with anti-NFAT5 antibodies. Immunoprecipitated chromatin was purified and sequenced. Chromatin aliquots from the wild-type and NFAT5-deficient BMDMs sample were separated to use as a measure of chromatin input. Libraries were prepared using the NEBNext Ultra DNA Library Prep Kit for Illumina (New England Biolabs, Ref. E7370) according to the manufacturer's protocol from 5-10 ng of starting material. Final libraries were analyzed using the Agilent Bioanalyzer with a DNA High Sensitivity chip to estimate the quantity and check size distribution, and were then quantified by qPCR using the KAPA Library Quantification Kit (KapaBiosystems, Ref. KK4835) prior to sequencing with Illumina HiSeq 2000
