Chromatin immune-precipitation was carried from 15 × 10^6 HDFs as for the ChIP assay with anti-AR antibody versus nonimmune IgGs. The bead-bound chromatin was tagged with Tn5 transposase (Nextera DNA Sample Prep kit, Illumina). The “tagmented” chromatin was de-cross-linked, purified, and amplified (20 cycles) using tag-specific primers. The amplified products were subjected to size selection (200-400 bp) and sequenced using Illumina Hi-seq.
