Lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody. Briefly, DNA was end-repaired using End-Repair Enzyme mix(Epicentre #ER0720) and dATP was added to 3’ends of DNA using Klenow fragment(NEB).After ligation of adaptors from Illumina Truseq DNA sample prep kit by T4 DNA ligase (NEB).DNA was PCR amplified with Illumina universal primers, library fragments near ~300bp were isolated from agarose gel and massive parallel sequencing was performed using Illumina HiSeq2000 sequencer.
