A total of 20 million cells per sample were collected, yielding ~20 fractions of chromatin. Chromatin was extracted using the MagnaChIP-seq kit (millipore).Sonication was performed for 12 cycles, 30 sec pulses using the biorupter Anti-histone H3 acetyl K27 antibody (ab4729, Abcam) antibody was used for ChIP. Per sample, 5–30 ng of precipitated DNA or input was used to perform library preparation according to the Illumina TruSeq DNA Sample preparation guide. In brief, the immunoprecipitated DNA was end-repaired, A-tailed and ligated to diluted sequencing adapters (1/100). After PCR amplification (15–18 cycles) and bead purification (Agencourt AmpureXp, Analis), the libraries were sequenced using the HiSeq 2500 (Illumina).
