Lysates were clarified from sonicated nuclei and H3.3-DNA complexes were isolated with antibody. Histone H3.3 chromatin immunoprecipitation was performed following the manufacturer instructions of Magna ChIP A Chromatin Immunoprecipitation Kit using 10 μg of Anti-Histone H3.3 antibody (Millipore). Sequencing libraries were prepared using the Kapa Hyper library Prep kit (Roche) with library amplification primer mix and sequenced as 50 base pair reads by single-read format on an Illumina HiSeq 2500 (v4 chemistry) with a maximum of 7 samples per lane
