Forelimbs from embryos at E11.5 were dissected and cells were disaggregated using a 2ml glass douncer. Nuclei were crosslinked with 1% formaldehyde for 5 minutes and quenched with Glycine (1mM final). Nuclear pellets were sonicated to a fragment range of 200-400bp using a Diagenode Bioruptor Sonicator. Sonicated extracts were precleared for 1 hour at 4˚C and incubated with anti-H3K27ac antibodies for 2 hours at 4˚C. Dynabeads (1:1 mixture of Protein A and G) were then incubated with the antibody-chromatin complexes for 30 minutes at 4˚C and ChIP-immune complexes were pulled down using a magnet.
