Phospho-H3Y41 Antibody Extraction Other: For treated samples, 20 Million cells were collected by centrifugation, resuspended in room temperature RPMI without FBS (2 Mln cells/ml) and cross-linked with 1% formaldehyde for 5 min at RT. Cross-linking was quenched by addition of 125 mM Glycine for 5 min at RT. Chromatin shearing was performed with a Bioruptor sonicating bath (Diagenode). Chromatin immunoprecipitation was performed with a phospho-H3Y41 antibody (Abcam #ab26310). ChIP-enriched DNA samples were used to create adapter-ligated libraries for massively parallel sequencing with the ChIP-Seq DNA Sample Prep Kit (Illumina).
