Irregular microscopic structures consisting of cords of endocrine cells that are scattered throughout the PANCREAS among the exocrine acini. Each islet is surrounded by connective tissue fibers and penetrated by a network of capillaries. There are four major cell types. The most abundant beta cells (50-80%) secrete INSULIN. Alpha cells (5-20%) secrete GLUCAGON. PP cells (10-35%) secrete PANCREATIC POLYPEPTIDE. Delta cells (~5%) secrete SOMATOSTATIN.

For RNA-Seq: Islets were enzymatically isolated from the pancreas and separated from residual exocrine debris. Isolated islets were dispersed into single-cell suspensions using trypsin. Lineage-tracing positive cells were enriched using MoFlo Astrios directly into TRIzol. Total RNA was extracted using RNAeasy Micro columns then DNase-treated with the TURBO DNA-free Kit and purified by RNAeasy Micro column. For ChIP-Seq: Islets were enzymatically isolated from the pancreas and separated from residual exocrine debris. Isolated islets were fixed and nuclear lysates were created. Chromatin was sheared and immunoprecipitated RNA-Seq Libraries: ScriptSeq Complete Gold – Low Input (Illumina, SCL6G); and ChIP-Seq Libraries: NEBNext Ultra DNA Libray Prep Kit for Illumina (NEB, E7370S)
