Bone marrow cells were extracted, red cell lysed and sorted for Lin-c-Kit+Sca1-CD34+CD16/32+ cells using a BD FACSAria. Lysates were isolated from sonicated nuclei and immunoprecipitation was done with an H3K27me3 antibody.
